Immunohistochemistry:AntibodyN52reactswithafragmentofNF-200side-armthatislocatedattheendoftheMPRKSPdomainandwhichcontainstheconsensuscdk-5phosphorylationsite,howeverthisreactivityisabolishediftheNF-200fragmentbecomesphosphorylatedbycdk-5{Guidatoetal,1996}.Thusforthefulleststainingandreactivity,includingwesterns,itissuggestedthatsamplesbetreatedwithalkalinephosphatasepriortoantibodystaining.Thefollowingtreatmentprotocolissuggested:
ProtocolPreparationofSolutions:
I.TBS:Tris-HCl,0.1mol/l;NaCl,0.1mol/l"pH8.0.
II.Mixalkalinephosphatase,1mg/mlwith1mol/lZnSO4,;1mol/lMgCl2,and1mmol/lPMSF;anddissoveinTBS.
NOTE:ItisnecessarytodialyzethealkalinephosphataseagainstsolutionIbeforeuse.
Procedure:
Frozensectionsfromshockfrozentissuesamplesareair-driedandthenfixedwithacetonefor10minat-20°C.Theexcessacetoneisallowedtoevaporateatroomtemperature.IncubatethesectionsinsolutionsIIfor4hoursat+30oCandwashinTBS3timesfor5minuteseach.NegativecontrolsectionsareincubatedinsolutionIIwithoutalkalinephosphatase.Furthertreatmentasfollows:
-Coverthepreparationwith10-20μlantibodysolutionandincubatefor1hrinahumidchamber.
-ImmersetheslideinTBSandwashinTBS3timesfor5minuteseach.
-Coverthepreparationwith10-20μlofasolutionofanti-mouseIg-antibody,labeledwithfluorscineisothiocyanate,andincubateinahumidchamberat37°Cfor1hour.
-WashtheslideinTBS3timesfor5mineach. |
