      HisTrap™FFcrudecolumnsoffertheconvenienceofsimple,one-steppurificationofhistidine-taggedproteinsdirectlyfromhomogenized,unclarifiedcelllysatebyimmobilizedmetalionaffinitychromatography(IMAC).

      HisTrap™FFcrudeisaready-to-usecolumn,prepackedwithprechargedNiSepharose®6FastFlow.Thisprepackedcolumnisintendedforpurificationofhistidine-taggedrecombinantproteinsbyimmobilizedmetalaffinitychromatography(IMAC).Afterthoroughcelldisruption,itispossibletoloadtheunclarifiedlysateonthecolumnwithoutprecentrifugationandfiltrationofthesample.Extendingthedurationofthemechanicaltreatmentofthesampletoensureamorecompletelysisisrecommended.Foroptimalresults,werecommendfirstadditionoflysozymeandDNaseIfollowedbyamechanicallysis,forexamplebysonication.Thehomogenizedsamplecanthenbeloadeddirectlyonthecolumnwithoutaclarificationstepwhichmaypreventdegradationofthetargetproteinandincreasetheactivity.
