Ac-Val-Gln-Val-Asp-pNA is a colorimetric Caspase 3 substrate. Caspase 3 is a cysteine protease that is rapidly activated during apoptosis and that cleaves several substrates, including poly(ADP-ribose) polymerase (PARP). Caspase 3 is activated by Caspases 8, 9 and 10 and cleaves and activates caspase 6 and 7. Caspase 3 cleaves the amyloid precursor protein (APP) contributing to Amyloid beta formation and has been implicated in Alzheimer’s disease and proposed as a target for drug discovery.
Activity is quantified by the release of p-nitroaniline which is measured by absorbance at 405 nm.
