Ac-Tyr-Glu-Val-Asp-AMC is a fluorogenic substrate for Caspase 1. Caspase 1/Interleukin 1b converting enzyme (ICE) is a cysteine protease that cleaves specifically at aspartic acid. ICE converts inactive IL-1 beta precursor to a pro-inflammatory cytokine that mediates a wide range of immune and inflammatory response. ICE-related cysteine proteases are required for E1A-induced, p53-dependent apoptosis in baby rat kidney (BRK) cells. Adenovirus E1B 19K protein, which is a potent inhibitor of apoptosis, inhibits activation of these proteases in BRK cells.
Activity is quantified by release of free fluorescent 7-amino-4-methylcoumarin (AMC) which excites at 360-380 nm and emits at 440-460 nm.
